Entwicklung von molekularen Werkzeugen zur Manipulation und Untersuchung von Gedächtnis-Engrammen

Das C01-Projekt bietet maßgeschneiderte optogenetische, virale und transkriptomische Analyseinstrumente zur Untersuchung von Mechanismen der Gedächtniskonsolidierung. Wir werden optogenetische Sensoren und Aktuatoren zur Kontrolle biochemischer Signalwege entwickeln und implementieren. Zur Optimierung des viralen Gentransfers und der Netzwerkanalyse verbessern wir bestehende retrograde und anterograde Tracer, und entwickeln neuartige Tracer auf Basis des Herpesvirus. Schließlich unterstützen wir die Anwendung von verschiedenen Einzelzell-omics-Plattformen, um zelluläre Komponenten von Gedächtnis-Engrammen zu identifizieren und involvierte Genexpressionsmuster zu untersuchen. Unsere Werkzeuge werden wir weltweit über DNA-Repositorien und unsere virale Core Facility zur Verfügung stellen.

Graphical Abstract: WP1 will customize and validate existing optogenetic tools and develop two new classes of optogenetic actuators that allow for light-gated Ca2+ influx while avoiding strong membrane depolarization, and optical bidirectional control of intracellular G-proteins and 2nd messengers. WP2 will develop viral systems for anterograde and retrograde trans-synaptic tracing for neuronal circuit analysis. WP3 will provide analytical tools for deciphering the transcriptional landscape of single cells at high-resolution. Collaborations for the WPs are indicated on the right.
