Modulation of engram cells by sharp wave ripples consequences for memory consolidation

Hippocampal sharp-wave ripples (SWRs) coordinate neuronal ensembles during memory consolidation, but how they engage experience-recruited inhibitory neurons remains unclear. Combining cFos tagging with _in vivo_ optotagging, we identified excitatory and inhibitory CA1 neuron recruited during spatial exploration. Tagged place cells were reused across contexts while maintaining distinct spatial maps. During subsequent rest, tagged place cells and interneurons showed enhanced SWR recruitment and preferential coupling.Anatomical analyses revealed enrichment of specific classes of interneurons within the tagged population. These findings identify inhibitory neurons as active components of experience-dependen ensembles coordinated during SWRs.This framework provides a basis for investigating memory dysfunction in Alzheimer’s disease. Our work in disease models shows also that memory impairment can accompany disrupted neuronal timing despite largely preserved place fields, highlighting the importance of coordination beyond spatial selectivity.

About the Speaker
Work in the lab of Silvia Viana da Silva “aims to further the current under- standing of the relationship between synaptic plasticity and entorhinal-hippocampal oscillations. We use state-of-the-art in vivo electrophysiology combined with novel imaging techniques to identify altered neuronal circuits in animal disease models. We believe our line of research can provide new ideas to guide therapies with the potential to ameliorate or restore the memory impairments characteristic of patients with Alzheimer’s disease of associated pathologies.” (Source (DZNE, AG Viana da Silva)
